Scampato a un'avventura nella foresta amazzonica, Indiana Jones, archeologo e avventuriero, riparte per cercare l'Arca dell'Alleanza di Mosè, sulle cui tracce si trovano anche gli emissari di Hitler (siamo nel 1936). Arrivato in Egitto trova l'Arca perduta in un pozzo popolato da serpenti, ma viene scoperto dai tedeschi che si impossessano dell'Arca e murano Jones dentro al pozzo. Indiana scappa, si mette all'inseguimento dei tedeschi e, dopo varie peripezie, li raggiunge nell'isola segreta in cui si sta per aprire il mitico contenitore.
